Single-cell analysis often focus on variation across clusters.
◼️ Yet, variation within clusters (conditions) is informative.

Proteins that do not change across conditions covary within a condition across single cells.

When is single-cell analysis needed ?

Should you measure nucleic acids, proteins or metabolites?

How do you achieve robust results ?

The regulatory capacity of dynamic systems is rich & fascinating.

They can enable a common regulatory hub to differentially regulate diverse physiological processes.

Do you remember the big questions that inspired you to become a scientist ?

Keep raising them.
◼️ Technological progress can convert scientifically intractable questions into tractable ones; they can be rigorously framed and empirically answered.

I still remember vividly the talks by Steve McCarroll and Ralph Weissleder.

Ralph presented a sweeping panorama of problems needing single-cell proteomics.

We first met in June 2018.
Since then, every meeting has reflected the annual progress of our community.

Based on the abstracts, 2024 will not disappoint.

I can barely wait for my teams to share results from Slavov Laboratory and Parallel Squared Technology Institute (PTI).

Nanopore direct RNA seq & siRNA knockdowns help identify pseudouridine sites and their synthase enzymes.

Well done Sara Rouhanifard and team!

We are yet to investigate proteomes with unbiased discovery-driven methods.

Yet, the prospects are good.
We may soon enter an age of discovery when proteomes are routinely explored at scale and their secrets revealed.

Let's elevate our scientific discourse:

◼️ Replace adjectives with factual statements
◼️ Offer constructive criticism
◼️ Place results in context where they can be better appreciated
◼️ Make research results accessible to broader audiences

Good point.
How should we name new phenomena that we do not yet understand ?

I favor phenomenological terminology describing the measurements; that's what we have used.

E.g., introducing high oxygen consumption (HOC) helped.
